How to Split Your Culture

How to Split Your Culture

  • Properly clean and sterilize 2 containers to be used only for culturing.
  • Remove the culture container lid, rigid airline tube and flexible airline tubing and place into a sink to clean for the culture that you are splitting. While at the sink, clean a funnel with Ethyl Alcohol and RODI water. Be sure to rinse the funnel until the Ethyl Alcohol smell is gone
  • Place the funnel into the first clean container; carefully without splashing,  pour 50% of the culture into the new container just below the middle of the container. Pour the remaining culture into the second clean container.
  • Using RODI water and a clean towel, wipe the outsides of the new container clean.
  • Pour new saltwater into the first new container using the funnel so that there are no spills. Do the same for the second container
  • Add F2 Media to the vessel. See the bottle’s instructions for the proper dosing amount.
  • Clean the outside and inside of a culture lids, rigid airline tubes and flexible airline tubing with Ethyl Alcohol and RODI water. Be sure to rinse the equipment until the Ethyl Alcohol smell is gone. Hook each culture up to their air pump. Remember do not cross contaminate

Take the original culture container and fill the bottom with RODI water. Using a dedicated bottle brush, clean the inside surface of the entire container with the brush. Rinse out with RODI water, empty it and allow it to dry in a sterilized, clean area. Spray the brush with Ethyl Alcohol, rinse with RODI water and allow to dry
